Abstract
This chapter reviews techniques used in isolating various pathogens of reptiles. The quality of the collected sample and the appropriateness of the procedure adopted to obtain it influence the entire isolation process. Fundamental considerations must be kept in mind when collecting specimens. Clinical and necropsy samples are often shipped to a diagnostic laboratory. Precautions need to be taken to ensure optimal preservation of samples prior to shipment, and overnight delivery is ideal. When shipping hazardous material, such as potentially infectious samples, extra safety precautions are required. Viral isolation requires the use of cell cultures or embryonated eggs, since viruses require live cells to replicate. Primary cell cultures are derived from fresh tissues that are treated with a proteolytic enzyme such as trypsin and are often composed of multiple cell types. Experience is very important and consulting personnel with extensive training and experience in cell culture and pathogen isolation is strongly advised.
